چکیده
I In a previous publication,8 we reported for the older Type II Sigma ribonuclease a value of An/c = 0.185 ml/gm. We now correct this value to 0.187 ml/gm; however, the precision cannot be evaluated with confidence. 8 Clarke, A. M., D. W. Kupke, and J. W. Beams, J. Phys. Chem., 67, 929 (1963). 9 McMeekin, T. Detection of proteins synthesized during the establishment of lysogeny with phage P22.
A method is described which utilizes pulse labeling with Sa5, starch gel electrophoresis, and autoradiography to detect the synthesis of proteins at various times after infection of Salmonella typhimurium with phage P22. The system was developed especially to study proteins involved in lysogeny. A comparison was made of various infections. These involve the wild-type phage (c+) under conditions...
متن کاملDual control of lysogeny by bacteriophage P22: an antirepressor locus and its controlling elements.
Two distantly linked clusters of genes on the Salmonella typhimurium phage P22 chromosome are involved in the control of lysogeny and superinfection immunity. One cluster consists of genes cl, ~2, and ~3, which are primarily responsible for the establishment and maintenance of lysogeny. It has been proposed that the second cluster consists of three loci which are responsible for the synthesis a...
متن کاملThe Synthesis of Phage and Host Dna in the Establishment of Lysogeny.
A method of extraction is described which prevents excessive fragmentation of bacterial DNA. The larger bacterial DNA can then be separated from the phage DNA on sucrose gradients. The relative amounts of phage and host DNA made at various times during infection of Salmonella typhimurium with phage P22 leading to lysogeny have been determined. Phage-specific DNA synthesis begins at about 4 minu...
متن کاملSynthesis and maturation of phage P22 DNA. II. Properties of temperature-sensitive phage mutants defective in DNA metabolism.
Three temperature-sensitive mutants of phage P22 have been isolated which are defective in phage DNA metabolism. After infection at restrictive temperature, one mutant (ts 12.1) fails to synthesize measurable quantities of phage DNA; nevertheless, parental phage DNA is recovered in a fast-sedimenting form similar to the first intermediate form observed after infection with normal phage. Another...
متن کاملControl of lysogenization by phage P22. II. Mutations (clyA) in the cl gene that cause increased lysogenization.
(Received 10 NovembPr IYSO, and in revised form 24 April 19X1) p12 cly mutants lysogenize at very high frequency after infection of wild-type Salmonella. One class of P22 cly mutations, clyA mutations, are shown to map within the P22 cl gene and to increase the activity of t,he cl gene product during establishment of lysogeny. The P22 clyil mutations also appear to affect c%-repressor gene expr...
